Product Application of  (2S)-1-(Chloroacetyl)-2-pyrrolidinecarbonitrile CAS#207557-35-5 

(2S)-N-chloroacetyl-2-cyano tetrahydropyrrole is a synthetic intermediate of vildagliptin, and is mainly used for the synthesis of vildagliptin. For example, L-prolineamide is used as a raw material, THF is used as a solvent, and K2CO3 is used as an acid-binding agent to carry out a chloroacetylation reaction to obtain (S)-1-(2-chloroacetyl)-2-pyridine carboxamide, and then the amide group is converted into a nitrile by a dehydration cyanation reaction with TFAA to obtain an intermediate (2S)-N-chloroacetyl-2-cyano tetrahydropyrrole. Finally, DCM is used as a solvent, K2CO3 is used as an acid-binding agent, and a condensation reaction is carried out with 3-amino-1-adamantanol, and then column chromatography is used for purification to obtain vildagliptin.
